HPLC purity measures what share of the peptide material in a vial is the correct target sequence, while net peptide content (NPC) measures how much of the total powder weight is peptide at all. They are two independent measurements, so a batch can show high HPLC purity alongside a much lower NPC.Lyophilised peptide powder also contains water, residual solvents and counter-ions such as TFA or acetate salts. NPC is determined by amino acid analysis or UV spectrophotometry and typically falls between 50% and 90%, depending on the sequence and the synthesis and purification method. For laboratory research use only, Daxer Labs advises researchers to check both values on the batch Certificate of Analysis (CoA) where reported, because calculating solution concentration from gross powder weight alone overstates the amount of peptide present.Read more: How to Read a Peptide Certificate of Analysis (COA): A UK Researcher's Guide (2026)
